Design: Samples were collected in duplicate from sexually-mature freely-mated adults (4-5 weeks post eclosion) D. grimshawi (G1 strain; Drosophila Species Stock Center ID: 15287-2541.00), the strain used for genomic sequencing and assembly (Drosophila 12 Genomes Consortium et al., 2007). Each sample is one individual female or male. We isolated total RNA and used polyA+ RNA for stranded PacBio RS II RNA-seq.
